Ketazol
Ketazol is used in the treatment of fungal infection by sensitive organisms
• Shampoo
• Cream
• Tablets
Indications
Limited to fungal infection by sensitive organisms. Tablets :
•
Systemic or visceral infections.
•
Cutaneomucosal infections where these infections cannot be treated due to the extent of the lesions or in case of resistance to the usual antifungal treatments.
•
Prevention of fungal infections in patients with congenital or acquired immune depression. Cream : Candidiasis,dermatophytes,pityriasis versicolor, erythrasma
Pharmacological Class
Imidazole derivative.
Pharmacological Properties
Antifungal.
Mechanism of Action
Operates at several levels in organisms
•
Membrane (increased permeability)
•
Cytoplasm (inhibition of oxidation processes in the mitochondria)
•
Nucleus (inhibition of RNA synthesis)
Side Effects
• Hepatotoxicity
•
Nausea, vomiting, diarrhoea
•
Pruritus, skin rash, urticaria.
•
Headache, dizziness.
Contradictions
Absolute : Allergy to ketoconazole and antifungals of the imidazole group.
•
Alcohol, midazolam (interaction)
•
Interaction : astemizole, cisapride, terfenadine, ebastine, triazolam.Relative :
•
Pregnancy or lactation
Observations
• The dosage specified above should be continued without interruption for at least one week once the symptoms have disappeared and until all the cultures prove negative
•
The penetration of ketoconazole into the CSF and brain tissues is low. Consequently, ketoconazole can not be used to treat meningitis
•
The use of antacids should be delayed by 2 hours (risk of reduced absorption of ketoconazole)
•
Patients who have exhibited allergic or hypersensitivity reactions after topical administration of imidazole derivatives should not be treated with oral ketoconazole
